两株产紫杉醇内生真菌J166和J137的
形态分类鉴定
摘 要
紫杉醇是近些年来开始使用的一种新型抗癌药物,对乳腺癌、肝癌等都有特殊的疗效.目前紫杉醇主要是从红豆杉等植物的树皮中提取,但其含量甚微,不能够满足人们对紫杉醇的需要。为了保护红豆杉同时解决紫杉醇的来源问题,本文对已分离出的产紫杉醇的红豆杉内生真菌进行形态鉴定,以便进行产紫杉醇内生菌的选育。为了鉴定从红豆杉植株中分离出的产紫杉醇内生真菌的菌种形态,本文对已收集到的太行山的红豆杉内生菌菌种进行了实验室接种培养和观察,对低温保藏的产紫杉醇内生真菌菌株进行筛选,挑选出无污染且菌落长势好的菌种作为接种菌,在无菌条件下进行接种,并置于人工气候培养箱中培养,定期观察菌落长势。待菌株产孢后对菌种进行光镜观察和拍摄,根据其特征、产孢结构、菌丝形态和分生孢子梗着生情况,对照真菌形态分类系统,对两个菌株进行初步鉴定。结果表明:得到的菌种J166初步鉴定属于半知菌亚门(Fungi Imperfecti)丛梗孢目(Moniliales)卵形孢霉属(Oospora Wallr.);J137初步鉴定属于半知菌亚门(Fungi Imperfecti)黑盘孢目(Melanconiales)粘盘孢属(Myxosporium Link ex Corda)。

The morphological identification of Producing-Taxol Endophytic fungus J166 And J137
Abstract
Paclitaxel is a new type of anti-cancer drugs start to using in recent years , it has a special effect on breast cancer ,liver cancer and other. At the moment paclitaxel is mainly extracted from the bark of the yew, but its content can't satisfy people's needs. In order to protect the fields and solve the problem of sources of taxol at the same time, we identify the producing-taxol endophytic fungus isolated from Chinese yew, in order to conduct breeding. This paper has inoculated and did laboratory training in the endophyte strains of Chinese yew of Taihang Mountains in China. The cryopreservation of taxol-producing endophytic fungi strains were chosen, in order to pick out the Pollution-free and colony growth good bacteria as inoculants, then inoculated the inoculants in sterile conditions, and placed it in the artificial climate incubator, observe the growth of the colony regularly. When they growed to a certain extent, observed their characteristics by optical microscope, In accordance with the classification of Endophytic fungus, study the hyphal morphology and terrier of the hygienic conditions of conidia than the classification system for the test to determine the species status of fungus The identification result of the two strains show that: Bacteria J166 belonging to Oospora Wallr. of Fungi Imperfecti Moniliaceae, bacteria L37 belonging to Myxosporium Link ex Corda of Fungi Imperfecti Melanconiales.
